Grow C. graminicola in Potato Dextrose Agar media for 2-3 weeks.
- Sterilize the surface of the seeds by following the Corn Seed Sterilization protocol.
- Wash vermiculite by placing it in a container for 2½ hours then pour out the water and autoclave for two hours. Allow the soil to cool.
- Fill 50ml tubes with 40ml of vermiculite.
- Cut plaques out of the 2-3 week culture of C. graminicola and place a layer of these over the soil.
- Add 5ml of soil to the tubes then place the seed in the tubes.
- Cover the seed with 5ml of vermiculite.
- Seal the top of the tubes using parafilm paper.
- Grow the plants under direct light at 28° C.
- After 3-4 days, as soon as the plants start to hit the parafilm, make a hole in the parafilm seal.
- Plants should be ready for observation 19-21 days after planting.
